He said that Mrs. Petrelli is 'playing' them so he knows that something is up.

It refers to the company policy of one special with one normal person. One of "us" (humans) one of "them" (post humans)

The phrase goes back to season 2. It's the protocol when partnering company members: One of us (normal human), One of them (evolved human).
I don't know about HRG and Mama P, though...He certainly knows her, and that she's manipulative.

Yeah...it's even mentioned in the GN where Sylar takes out the two agents who are in love...he mentions this theory...personally, I'd want the Haitian...much safer lol

When I first heard Noah say "one of us, one of them" I believed he was grouping himself with Angela Petrelli. Everyone knows that they pair one human with someone with powers. However, it seems that there is much more significance when he said it to Mrs. Petrelli, after all they did name the episode after it. I have been racking my brain on what connection he must have with Angela Petrelli and have a few wacky theories.
But, if it didn't mean anything... they should have re-worded it to make more sense considering Angela Petrelli does have a power and wouldn't be considered an "us" but a "them".
But I also like the theory :: one of us (Good Guy), one of them (Bad Guy)... that makes sense.

I think the deeper meaning of the show's title was being hinted at in the intro - "What kind of person are we? Does the hero, or the villain inside us, win the day?"
This is "one of us and one of them" the hero and the villian in each of us.
So we have:
Noah & Sylar = one non-powered & one powered, one "goodie" and one "baddy"
Nathan - battling with internal angels and demons
Peter/Jesse = one good one bad - the hero inside the villain
Sandra & Meredith = Claire's mothers: one non-powered & one powered
Seems like a pretty apt title to me...

Yes...
The double meanings of the titles on this show always make me happy.

SamuraiSword,
When talking about the Company's "normal" two-person field agent teams, I think the "us" refers to a super-powered human (HEB) and the "them" refers to a non-powered human (I imagine this to be more correct from Angela's perspective). [NOTE: Of course, nothing changes when viewed from the us being a non-powered human perspective.]
I don't think they were referring to one hero and one villain.
